STUDY OF VARIABILITY AND RESPONSES TO STRESS IN OAK BY A MULTI-omic APPROACH (transcriptomics, proteomics and metabolomics). ENCIN-OMICA

 

We aim at incorporating DNA molecular-markers and modern –omics (transcriptomics, proteomics and metabolomics) techniques to the study of population variability, development processes (maturation and germination of seeds) and response to stresses (drought and Phytophthora cinnamomi) in holm oak (Quercus ilex). Despite its environmental (predominant species of the Mediterranean-forest ecosystem) and economic interest (part of “dehesa”), the holm oak is an orphan species, with molecular studies almost absent. The project has been structured into eight objectives: i) Survey of twenty geographical locations all over Andalusia; ii) Studies of the variability of response to drought and Phytophthora cinnamomi among the 20 populations; iii) Analyses of population variability by using chloroplastic SSR markers (chloroplast microsatellites); iv) Metabolomic analyses of acorns and leaves; v) - vi) Proteomics and transcriptomics analyses of the response to drought and Phytophthora cinnamomi; vii) Proteomics analyses of seed maturation and germination; and viii) Integration of the data and construction of a holm-oak database. The proposal is a follow up of a project initiated 12 years ago by two research groups at the University of Cordoba: "Agroforestry and Plant Biochemistry and Proteomics" and "Restoration of Agricultural and Forestry Areas”. It incorporates the “Agrifood Biotechnology" one. These groups have experience and international prestige in the proposed topics and methodologies. To date, our groups have made a major effort in developing molecular  techniques to study the forest-tree biology, being species of great difficulty as experimental systems. Our research within this field has been very productive from scientific, translational and academic points of view, favoring the integration with international groups. This proposal aims to leap further in the direction of systems biology, implementing the techniques widely  used in our laboratories (forestry, remote sensing, ecophysiology, microscopy, classical biochemistry, NIRS and proteomics) with those of DNA markers (microsatellites), transcriptomics, metabolomics and bioinformatics. The proposal aims at generating knowledge and methodologies to characterize biodiversity within this species and to establish the phenotypic and molecular basis for the selection of elite individuals, those presenting the highest adaptability and resistance to stresses, such as those associated with the decline syndrome or predicted in a scenario of climate change. Because of that, it will be translated to the forestry sector to assist reforestation, conservation and sustainable management practices of forest ecosystems. There are similar programs   worldwide, but none of them include holm oak, as it is a typical species of the Mediterranean basin; specifically in Spain.   The project fits within the scientific standards required by the Spanish Science and Technology system regarding: internationalization (involvement of two European groups), publications, impact, environmental and social concern; as well as support and active participation of PRBs and multidisciplinarity. We also pretend to translate it to the academia, as a way to promote forestry research in degree, master and thesis programs. It will help to strengthen international partnerships and initiatives in forestry, particularly with Hispanic America and North Africa, being areas with which our groups have already initiated similar projects.
